چکیده

To determine whether release of tumor necrosis factor-a (TNF-a), a cytokine that affects iron homeostasis, may be selectively altered in hereditary hemochromatosis, we measured concentrations of TNF-a and interleukin-ls (IL-ls) in supernatants of cultured peripheral blood monocytes from 11 homozygotes for hereditary hemochromatosis, 11 healthy individuals, and five patients with iron-loading anemia. The gene for hereditary hemochromatosis is tightly linked to the HLA locus on chromosome 6, but its exact site and product are not known. The gene for TNF-a also is located within the HLA region. Monocytes were incubated from 4 to 36 hours in medium alone or with added lipopolysaccharide. Mean concentrations of immunoreactive TNF-a in supernatants were

Decreased concentrations of tumor necrosis factor-alpha in supernatants of monocytes from homozygotes for hereditary hemochromatosis.

To determine whether release of tumor necrosis factor-alpha (TNF-alpha), a cytokine that affects iron homeostasis, may be selectively altered in hereditary hemochromatosis, we measured concentrations of TNF-alpha and interleukin-1 beta (IL-1 beta) in supernatants of cultured peripheral blood monocytes from 11 homozygotes for hereditary hemochromatosis, 11 healthy individuals, and five patients ...

مقایسه تأثیر Tumor Necrosis Factor-α، Monocyte Conditioned Medium و polyinosinic-polycytidylic acid در بلوغ سلول‌های دندریتیک

Abstract Background and purpose: Different laboratories have considered different maturation factors for producing human dendritic cells (DC) from peripheral blood monocytes. In this study, we comprehensively investigated the effect of adding poly (I:C) to standard maturation stimulus, i.e. MCM and TNF-α on the induction of T cell response. Materials and methods: Peripheral blood monocytes ...

Tumor necrosis factor alpha promoter polymorphisms influence the phenotypic expression of hereditary hemochromatosis.

Severe iron overload usually develops in patients with hereditary hemochromatosis (HHC), but variability in the phenotypic expression of the disease has been reported. This study assessed whether tumor necrosis factor alpha (TNF-alpha) plays a role in phenotypic expression of HHC. Sixty-four patients with HHC and 172 healthy volunteers (controls) were studied. Release of TNF-alpha from stimulat...

Alterations in Gene Expression of Interferon and Tumor Necrosis Factor‐α in Human Blood Macrophage-Like Monocytes Induced by Clinical and Standard Salmonella typhi Strains in vitro

Background and Objective: Salmonella typhi as a human pathogen stimulates the human immune system and triggers gene expression changing its pathogenesis. Therefore, we aimed to investigate the expression levels of ifn-γ and tnf-α cytokines in human blood macrophage-like monocytes in dealing with clinical and standard samples of Salmonella typhi in vivo. Materials and Methods: In this cross-sec...

Diminished transcription of interleukin-8 by monocytes from preterm neonates.

Developmental delays, which impair antibacterial host defense, are present in the neutrophil system of human preterm neonates. We hypothesized that diminished production of interleukin-8 (IL-8), a neutrophil chemotactic peptide, might in part be responsible for these defects. To test this, monocytes from the blood of preterm neonates, term neonates, and adults were isolated immunologically by "...
